Born Mark Isaakovich Prudkin on September 14, 1898, in Klin, Russian Empire, he was the son of Isaac Prudkin, a tailor. Prudkin studied acting at the 2nd Studio of the Moscow Art Theatre from 1918 to 1924. He joined the famed Moscow Art Theatre troupe in 1924, invited by Konstantin Stanislavski.

Though Prudkin made a brief film debut in 1927, he chose not to act in movies for the next 40 years, believing that stage acting was superior. He finally returned to film in 1969 with a noteworthy role as Fyodor Karamazov in The Brothers Karamazov, directed by Ivan Pyrev, Kirill Lavrov, and Mikhail Ulyanov.
